   Research Interests:

1. Anti-HIV-1 drugs screening and the structure-activity relationship study of lead compounds  
An anti-HIV drug screening platform has been established, which includes primary screening, confirmation and specific research at the levels of molecular, cellular and AIDS primate animal models in our lab. We have widely collaborated in the study of anti-HIV drugs with 100 units from 24 provinces in China, and 6 overseas agencies from Russia, India, etc. More than 10,000 compounds purified from animals, plants, and halobios were screened for their anti-HIV-1 activities, and a series of compounds with anti-HIV-1 activities (e.g. Xanthohumol, Concentricolide and Schisandrins) were found. We have completed preclinical anti-HIV activities evaluation for 11 anti-HIV drug candidates. TCS recombinant mutant (Approval No: 2005L00148)Sifuvirtide (Approval No: 2005L01088)and Azvudine (Approval No: 2013L00941) has won the drug clinical trial approval document issued by CFDA. “Tangcao Pian” is the first and unique licensed TCM for AIDS in China, the Sifuvirtide recently entered phase III clinical trials in HIV-infected individuals, the Azfudine (FNC) and mTCS completed phase I clinical trial. Till now, 49 patents applications have been granted and filed. More than 150 research papers in anti-HIV drugs have been published. Our lab has become one of the main groups of anti-HIV drug research in the country now.  

2. SIV/SHIV/HIV non-human primate models for AIDS, host restriction factor, and the pathogenic mechanism of AIDS 
We have established five SIV/SHIV viruses ( including SIVmac239-, SIVmac251-, SHIV89.6-, SHIVSF162P3-, and SHIVwhu-.) infected Chinese rhesus monkey (Ch Rhs) models. In SIV/SHIV infected monkey models, our interests mainly focus on the functions of DC and Treg cells in the process, especially the early stage of simian AIDS, and the roles of old age, virus induced immune activation in disease progression. We have previously reported Northern pig-tailed macaques (Macaca leonine, NPMs) were susceptible to HIV-1 in vitro due to absence of a TRIM5 restricting protein, suggesting NPMs might be a potent animal model for HIV infection. We infected 4 Northern pig-tailed macaques with a HIV-1NL4.3 strain, and our data demonstrated HIV-1 infection was established and viruses were latent in all 4 animals. The HIV-1/HSIV infected northern pig-tailed macaques are in progress. Besides, we identified the TRIM5/TRIMCyp heterozygous genotype in M. assamensis. Recently, we found a novel mode of CypA insertion in tree shrew TRIMCyp gene fusion (ts TRIMCyp). tsTRIMCyp lost restriction ability against retroviruses and play an important role in regulating NF-κB and IFN-β promoter activation as a negative regulator. tsTRIMCyp may play a role in preventing over-reaction of innate immune response and maintaining immune balance 

3. The HIV/AIDS epidemic in Yunnan province and transmission in southwest China 
The China-Myanmar border is a particularly interesting region that has very high prevalence of and considerable diversity of HIV-1 recombinants. We found that Northern Myanmar is a big forge for HIV-1 recombination between CRF01_AE, B’, and C because the extremely high proportion of intersubtype recombinants, especially CRF01_AE/B’/C recombinants (42.6%). We also firstly represented the molecular epidemiological characterization of HIV-1 among Burmese LDTDs. High prevalence of HIV-1 CRF01_AE and inter-subtype recombinants, as well as dual infection, were detected among them. Burmese LDTDs may have made contribution in HIV-1 transmission between the heterosexuals and IDUs and between China and Myanmar in general. Co-infection with HIV and HCV and/or HBV is highly prevalent among the IDUs in the China-Myanmar border region. All infections were more prevalent among the Chinese IDUs than among the Burmese. We have isolated 421 HIV-1 clinical strains from HIV-1-infected individual in Yunnan.
